泛素(Ubiquitin,Ub)是一種高度保守的小蛋白(76個氨基酸),普遍存在於從單細胞酵母到人類的所有真核細胞內,並且表達水平很高。通過一系列的酶促反應,泛素在泛素結合酶(E2或Ubc)和泛素連接酶(E3)的幫助下連接到目的蛋白。接著在一個被稱為多聚泛素鏈的泛素多聚體形成后,結合有多聚泛素鏈的目的蛋白被26S蛋白酶體降解,而這個過程是許多細胞活動調節的重要手段,2004年的諾貝爾獎授予了“泛素化的發現和它在蛋白降解中的作用”就說明了它的重要性。深入研究揭示,上述泛素鏈是由下一個泛素表面的Lys48殘基結合在上一個泛素的C末端甘氨酸殘基而形成的,隨後又發現泛素鏈也可經由表面Lys63殘基形成。在所有生物中發現的泛素結合酶中,只有Ubc13能夠專一通過Lys63連接泛素。Ubc13的獨特性質是基於它和另一個被稱作泛素結合酶異構體的蛋白的結合,而這種異構體也是Lys63多聚泛素化過程所必須的。更重要的是,被Lys63連接的泛素所結合的目的蛋白不是為了降解,而是為了改變其活性。此外,在其他泛素化類型中,單一泛素化也被認為是一種新穎的蛋白質活性調節機制,拓寬了人們對泛素化功能的認識。本實驗室的主要興趣在於發現上述非常規泛素化過程和探索相關途徑的分子機制。因為一些研究的很透徹的非常規泛素化目的蛋白參與一些重要的細胞代謝過程,如DNA損傷耐受、細胞周期檢驗點、自然免疫和應激反應,而且這些過程在人類疾病中起關鍵作用,如癌症,抗病及衰老,所以這項研究將會對包括癌症在內的多種疾病診斷和治療產生直接影響。
