研究內容


植物病原菌的特徵決定了植物對其響應(先天免疫)主要發生在兩個層面上。一個是對病菌表面保守的分子特徵(Pathogen Associated Molecular Patterns,PAMPs)的識別,如細菌的鞭毛蛋白Flagella和伸長因子EF-Tu。這個層面的識別可以啟動基於細胞膜上的受體或受體似蛋白的信號傳遞,從而激活抗性基因的表達(PAMPs triggered immunity, PTI)。另一個層面上是對病菌內的致病因子(effector)的識別。這個層面的識別會激活更強烈的反應,典型的反應是細胞編程性死亡(或稱“超敏反應”)。這一類的反應主要是抗性基因(R基因)的啟動,稱之為ETI (Effector triggered immunity, ETI)。
在眾多植物先天免疫模型中,植物膜蛋白RIN4和病菌的分子互作機制被研究最為透徹。在近期的研究中,我們發現了植物受體似激酶家族VII的一個蛋白RIPK可以特異性地磷酸化RIN4。磷酸化的RIN4可以被抗性基因RPM1識別從而啟動ETI。植物受體似激酶VII是一個大家族,由於其參與了植物先天免疫,我們將用酵母雙雜交和免疫共沉澱的辦法尋找這些植物中同源基因作用的蛋白,分析其在植物先天免疫中的作用。
由於病菌和植物互作主要發生在細胞質膜上,對病菌入侵信號識別的受體和受體似蛋白大多分佈在細胞質膜上,因此對膜蛋白及其結合蛋白的研究具有重要的意義。我們現有的技術可以獲得非常純的細胞質膜,結合先進的質譜技術,使得我們能夠鑒定和分析病菌侵染后膜蛋白組學的變化。通過對膜蛋白質組學的研究,揭示膜蛋白及其作用蛋白在植物抗病中的意義和機理。
