真核細胞發育和分化過程中,功能基因組的表達受到精細而廣泛的調控,主要通過相應的調控元件如啟動子增強子等實現。近年來的研究發現,真核細胞內染色體組蛋白的特定修飾與基因調控元件密切相關,如H3K4me3特異性標記啟動子,H3K4me特異性標記增強子,而H3K27ac標記活化的調控原件。通過組蛋白特定修飾相對應的抗體進行高通量的Chip-seq實驗,在全基因組範圍內揭示功能基因組的調控元件,並結合生物信息學的方法統計分析與調控元件相結合的轉錄因子,從而確定真核細胞發育和分化的關鍵基因及其調控網路。系統性的研究生命過程中功能基因組表達和相互之間的調控關係,從而繪製基因調控迴路,全面解析生命過程。
