1. 玉米分子遺傳學研究。主要針對玉米籽粒的蛋白品質性狀,開展重要突變體的基因克隆和功能分析。重點針對玉米籽粒Opaque突變體(為主要影響籽粒蛋白含量的一類突變體),開展圖位克隆和功能分析。並在此基礎上開展對玉米主要儲藏蛋白的轉錄調控機制研究。
2. 玉米基因組學研究。功能基因組學研究:利用玉米Ac轉座子系統,創建玉米功能基因組學研究的遺傳材料,並探索相應的研究方法。比較基因組學研究:開展玉米重要染色體位點的比較基因組學研究,特別關注玉米關鍵性狀的馴化問題。
以往主要研究基礎:
1. 開展了針對玉米重要性狀的分子遺傳學和基因組學研究,其中對玉米主要馴化基因tb1的馴化突變位點的研究發表在Cell Research(2011)。對玉米重要籽粒突變體Opaque7的圖位克隆研究發表在Genetics(2011),是國內首個發表的玉米圖位克隆研究工作。構建了國內首個可用於大規模玉米功能基因組研究的玉米Ac轉座子突變系統(Genetica,2010)。
2. 系統開展了針對玉米主要儲藏蛋白(醇溶蛋白)的基因組學、比較基因組學和功能基因組學研究,相應研究以第一作者發表了Genome Research (2001),Genome Research(2002),Plant Physiology(2002)和PNAS(2003);以通訊作者發表了Plant Mol Biology(2009),Plant Mol Biology(2010)和Genome(2010)。
3. 在玉米中最先嘗試了RNAi轉基因技術,並用單個RNAi構建沉默一個基因家族,以第二作者發表Genetics(2003);在玉米中嘗試了超大片段的遺傳轉化,也是首次完整轉化一個大型的簇狀基因家族(高梁22kD醇溶蛋白基因簇),以第一作者發表Nucleic Acids Research(2004)。實驗室建有成熟的玉米轉基因技術平台,承擔有轉基因育種國家重大專項任務。
4. 參與了國際水稻基因組計劃中第10號染色體的序列測定工作,具體負責了Rutgers大學分攤的區段,是Science(2003)和Nature(2005)的共同作者,並因此得到美國農業部頒發的獎勵。回國后還開展了對14個野生稻在Moc1位點的比較基因組學分析,以並列第一作者發表PNAS(2009)。
5. 開展了極端耐逆生物(鹽藻Dunaliella viridis和鹽芥Thellungiella halophila)的功能基因研究,報道了多個獨特功能基因的克隆和功能分析,以通訊作者發表Plant Molecular Biology等SCI論文10篇。
