他的研究領域為中樞神經系統突觸可塑性,近年來主要研究膠質細胞激活和致炎細胞因子過表達引起慢性疼痛、記憶障礙和抑鬱症的機制。他首次發現脊髓背角C纖維突觸長時程增強,現已被認為是病理性疼痛的突觸模型。他發現外周神經損傷后TNF-過表達通過上調DRG神經元的鈉通道和增加脊髓背角興奮性突觸聯繫引起慢性疼痛。TNF-還通過損害海馬的結構和功能導致記憶障礙。
長期從事病理性疼痛的中樞和外周機制的研究。在SCI收錄的期刊上發表二十餘篇研究論文,被引用近500次。主要學術成就包括:
(1)首次報道了脊髓背角C纖維誘發電位的長時程增強(LTP)。發現激動NMDA、NK1和NK2受體是誘導LTP的必要條件;自然痛刺激可引起C纖維誘發電位的LTP,但在正常情況下受到下行抑制系統的抑制。該LTP被認為是痛覺過敏的基礎;發現脊髓LTP的早期維持與PKA、PKC和CaMKII的激活有關,而脊髓LTP的晚期需要新的蛋白質合成;多巴胺D1/D5受體在LTP的晚期維持中起重要作用。
(2)發現電刺激Aδ纖維在正常動物引起C纖維誘發電位的LTD, 然而同樣的刺激在脊動物引起LTP, 證實下行抑制系統決定脊髓背角突觸傳遞可塑性變化的方向。
(4)發現運動神經(腰5前根)損傷引起同側背根神經節和雙側側脊髓背角TNF-a及其受體1(TNFR1)上調,並證實TNF-a參與病理性疼痛的形成而不參與其維持。在正常動物不同濃度(10pg/ml、4.5ng/ml和100ng/ml)的TNF-a不影響強直刺激引起的脊髓背角LTP,然而,在腰5前根引起痛覺過敏的動物,小劑量的TNF-a(10 pg/ml)就可引起脊髓背角C纖維誘發電位的LTP。TNF-a通過激活NFkB、JNK和P38而發揮作用。
